ABSTRACT

OBJECTIVE To explore the mechanism of Cirsium japonicum extract in improving hypercholesterolemia based on metabolomics technology. METHODS The extract of C. japonicum was prepared by macroporous resin adsorption, and its main components were identified by liquid chromatography-tandem mass spectrometry. The experimental mice were randomly divided into control group (n=6) and modeling group (n=16). The hypercholesterolemia model was induced by diet in modeling group; after modeling, the rats of modeling group were divided into model group (n=8) and C. japonicum extract group (n=8). C. japonicum extract group was given C. japonicum extract 400 mg/(kg·d) by gavage (calculated by extract), and other 2 groups were given constant volume of 0.3% sodium carboxymethyl cellulose solution, for 6 weeks. After medication, the intervention effect of C. japonicum extract was evaluated by the levels of serum total cholesterol (TC), triglyceride (TG) and the histopathological changes of liver. The mechanism of C. japonicum extract in improving hypercholesterolemia model mice was investigated by metabolomics. RESULTS It was identified that C. japonicum extract contained 12 components, such as 030302005) chlorogenic acid, linarin and pectolinarin. After 6 weeks of intervention, compared with control group, serum level of TC was increased significantly while the level of TG was decreased significantly in model group (P<0.05), while a large number of lipid droplets, disorderly arrangement of liver cells and the damaged structure of liver cord were observed in liver tissue. Compared with model group, the serum level of TC was decreased significantly in C. japonicum extract group(P<0.05); the lipid droplets in liver tissue were significantly reduced, with liver cells arranged radially and tightly centered around the central vein, and liver cords arranged neatly. The metabolomics study showed that after the intervention of C. japonicum extract, the levels of metabolites were significantly adjusted back, such as ethanolamine, fumaric acid and cholesterol; finally, three metabolism pathways, such as alanine-aspartate-glutamic acid metabolism, arginine biosynthesis, citric acid cycle, were obtained. CONCLUSIONS The main components of C. japonicum extract are phenolic acids and flavonoids, such as chlorogenic acid, linarin, pectolinarin. C. japonicum extract can improve hypercholesterolemia by regulating the contents and distribution of differential metabolites, adjusting alanine-aspartate-glutamic acid metabolism, arginine biosynthesis and citric acid cycle, participating in oxidation-reduction reaction, improving liver lipid accumulation, and playing anti-inflammatory role.

ABSTRACT

Objective To investigate the related factors affecting the efficacy of microvascular decompression (MVD) in the treatment of primary trigeminal neuralgia (TN). Methods MVD was performed in 120 patients with primary TN and single factor χ2 test and Logistic regression analysis were performed on the clinical data to evaluate the independent risk factors influencing the efficacy of MVD in the treatment of primary TN. Results The pain completely disappeared in 101 cases, the pain was relieved in 16 cases and the unhealed in 3 cases. Sixty cases had pure arterial compression,57 cases had venous compression, and 3 cases had no obvious responsibility vessel. Complete elimination of pain in the patients with venous compressionwaslower than that in patients with pure arterial oppression: 73.7% (42/57) vs. 86.7% (52/60), and the difference was statistically significant (P<0.05). The complete pain relief rate in patients with V2 bronchial pain was lower than that in other pain distribution areas: 13/19 vs. 87.1%(88/101), With significant difference (P<0.05). In patients with V2 bronchitis, the responsible vessels were mostly compressed from the ventral surface of the trigeminal nerve, and the difference was statistically significant (P<0.05). Logistic regression analysis showed , Venous compression and V2 pain were the independent risk factors influencing the clinical efficacy of MVD in the treatment of primary TN. Conclusions Patients with venous compression and V2 pain are less likely to havegood postoperative outcomes, and venous compression and V2 painare independent risk factors. In patients with V2 pain, the responsible vasculature tends to ventrally massage the trigeminal nerve, which is a new study direction of refractory V2 pain.

Objective]To research and explore the characteristic of medicine in Treatise on Febrile Diseases. [Methods]Combine with the doctors of past dynasties, relevant annotations of the treatment of cough notes in Treatise on Febrile Diseases,and my own learning experience,analyze the characteristic of medicine of Zhongjing treating cough in Treatise on Febrile Diseases.[Results]Its characteristics are summed up in three points.Pay attention to the thought of the simultaneous dispersion and descending, and conform to lung qi.Pay attention to the drugs of the lung channel,especially the application of Ephedra and its compatibility with other drugs.Pay attention to“reinforcing earth to generate metal”and protect stomach qi. [Conclusion]The research of this paper is beneficial to broaden the clinical treatment of cough ideas,expand the clinical application of the classical prescription,deepen the understanding of Zhang Zhongjing's academic thought.

ABSTRACT

To obtain a recombinant human plasminogen (hPLG) with potential anti-platelet aggregation activity, we cloned the cDNA coding Pro544 to Asn791 of hPLG, a kringle-deficit derivative (hPLG-deltaK). The Pro559 in activation loop was then mutated into Asp559 to provide Arg-Gly-Asp (RGD) motif. The constructed pPICZalphaA-RGD-HPLG-deltaK plasmid was expressed in yeast Pichia pastoris GS115, which produced RGD-hPLG-deltaK about 0.160 g/L broth. After affinity chromatography, the purity of the recombinant protein reached above 90%. Western blotting test confirmed that it retained the immunological reaction capability as human PLG. Its urokinase activation rate in 24 hours and its fibrinolytic activity made no deference against native hPLG-deltaK (P=0.630, n=5). Importantly, after activation by urokinase, RGD-hPLG-deltaK showed a significantly higher platelet aggregation inhibition rate (Ri) (21.8% +/- 1.57%) than hPLG-deltaK (3.8% +/- 0.33%) (P=0.000, n=5). These results proved that we constructed an hPLG mutant with anti-platelet aggregation activity, which made a foundation for developing innovative thrombolytic drugs with multifunction.

ABSTRACT

Dysentery is one of the digestive system diseases, which has a higher incidence rate. The aim of present study was to further explore the incidence of dysentery and get the better prevention of this disease. The data were collected and statistically analyzed from Shaanxi Province for the monthly incidence of dysentery in 22 years. In addition, according to the Yunqi theory and related incidence of diseases, the correlation between the incidences of dysentery and Suiyun, Sitian, Zaiquan and Six-qi were summarized systematically. This study provides a reference for the incidence of dysentery.
